Botanical blend BluNADBooster shown to increase and preserve NAD⁺ levels in healthy aging adults

A clinical trial examined the effect of the herbal supplement from NXT USA on supporting cognitive, physical, and inflammatory markers.

New research indicates supplementation with BluNADBooster both increases and preserves nicotinamide adenine dinucleotide (NAD⁺) levels, improving biomarkers of healthy aging.1 This herbal blend from NXT USA is made from standardized extracts of Punica granatum (pomegranate) and Tagetes erecta (marigold), and demonstrated a dual-action approach to NAD levels, a December 9 press release from NXT USA explains.2 The ingredient is also highlighted for its high polyphenol content.

Study design: measuring markers of aging

The randomized, double-blind, placebo-controlled clinical trial, “Comparative efficacy of a standardized herbal composition LN22199, nicotinamide riboside, and their combination on NAD+ metabolism in healthy aging adults,” was published in the Journal of Functional Foods in December 2025.

In this study, BluNADBooster (called LN22199) was compared to the NAD+ precursor nicotinamide riboside (NR), and a combination of the ingredients was also examined. The trial included 140 healthy and aging adults, who received either 1000 mg of LN22199, 500 mg of NR, a combination of 500 mg of LN22199 and 500 mg of NR, or a placebo for 60 days. The main measurements were for NAD+ levels, CD38 activity, inflammatory biomarkers, cognitive function, physical performance, and quality-of-life measures.

As the study explains, a depletion in NAD⁺ levels with aging contributes to mitochondrial dysfunction, inflammation, cognitive and physical decline, which is driven in part by cluster of differentiation 38 (CD38) activity.

Interest is increasing for plant-derived compounds for modulating NAD⁺, the study adds, as potentially safer and more sustainable options. Additionally, Punica granatum contains polyphenols that can provide antioxidant, anti-inflammatory, and anti-aging benefits, while Tagetes erectai has demonstrated hepatoprotective, neuroprotective, and ocular benefits in other studies.

Results: enhancing NAD+ metabolism and decreasing CD38 activity

LN22199 supplementation was shown to support increases in NAD+ metabolism as well as both cognitive and physical functions, while reducing CD38 activity and inflammation.

Highlighted results include:

  • Blood NAD⁺ increased with LN22199 alone increased by 26.48%
  • CD38 activity decreased by 17.98% with LN22199 alone
  • Serum growth/differentiation factor 15 (GDF-15) were reduced by 15.07% with LN22199

These results supported preclinical data that had suggested LN22199 synergistically promoted NAD+ synthesis and reduced CD38 activity, the researchers noted. “These findings support a dual-action strategy of boosting NAD+ biosynthesis and limiting its degradation as a promising approach to mitigate advancing age-associated decline,” they concluded. “LN22199, when used alone or in combination with NR, provides a comprehensive intervention strategy that emerges as a safe and effective nutraceutical approach to promote healthy aging and mitochondrial function.”

The treatments were described as well tolerated and no serious side effects were reported.

Impact for NAD+ supplementation

Blue Helix and NXT USA developed the blend of botanicals based on their synergistic potential, the press release explained.

This study confirms what we’ve believed for years: polyphenols matter, and they work in ways synthetic precursors cannot.”

The results and safety profile supported the use of the supplement for healthy aging benefits, the study noted.

“This is comprehensive, multidimensional evidence—science layered on science—to ensure the formulation truly delivers,” added Anderson. “For the first time, we’re seeing a clinically validated botanical strategy that supports multiple hallmarks of aging—including mitochondrial efficiency, inflammation, and cognitive resilience—within just 60 days.”
